You'll need to find the elevation for your location, a simple search on the web should get you there. On level ground first set the elevation, then the pitch and roll. that's about it.

It's just a simple barometer and not particularly accurate. The reading will change not only on altitude but also as the local atmospheric pressure changes with varying weather patterns.
If you really want to "calibrate" it then you need to dial it to your current elevation (just google elevation of city "X", with X being wherever you are) and do so on a day when there isn't a particularly high or low pressure system influencing the reading.
Don't expect this gauge to accurately tell you when you've gone up or down a 100' in elevation! However, if it reads 0 +/- a few hundred feet at the beach then it will read 6000' +/- a few hundred feet in Denver.

Yes, adjust it to a known elevation. I usually find with mine that I set it to the elevation signs along most of the major highways here in CA. If I set it once above 4000' when going up or set it once below 4000' going down, then it is fairly accurate. And with a big storm moving it, it'll read a few hundred feet high due to the low pressure of the storm.
