Talking Timing ? Stuck cylinder ? Throttle body backfire ? Need help

Having back fire through the throttle body, I’ve had a no start situation ion my 95 4Runner for
a week now, I also bought it a week ago and was told that the distributor was bad.. replaced it, it’s acting like it wants to start but doesn’t. Possibly 180 degrees out or 360 degrees out. I placed a long screwdriver in cylinder 1 and waited for it to move outward as I rotated the crankshaft 360 degrees multiple times... screw driver didn’t move. I was trying to find the compression stroke. Every fuse is great and are getting power, one bad relay to my knowledge and that’s the efi relay. I temped up the relay to make it work because it was making my diagnostic jumper not work at all, I also knew this because my CEL on the dash was not lit up. Needless to say I’m stumped but keep pressing forward to get this 4Runner back to life. To note: it’s getting fuel, spark, new spark plugs, new wires, ignition coil is getting power, igniter is getting power, and the throttle body is back firing.. im also getting code 41 TPS. Any help is greatly appreciated.
